Incucyte® + Octet® 双剑合璧

三阴性乳腺癌(Triple negative breast cancer, TNBC)是乳腺癌中最为棘手的一类。不断有研究表明,CDK12/13 是 TNBC 治疗的潜在治疗靶点。2022年8月发表在药物化学TOP期刊 Journal of Medicinal Chemistry 杂志上的一篇文章[1],通过采用蛋白水解靶向嵌合体 (PROTAC) 技术发现的第一系列高效和选择性双 CDK12/13 降解剂。其中最优化合物 7f  降解 CDK12 和 CDK13 的 DC50 值分别为 2.2nM 和 2.1 nM。化合物 7f 表现出对 CDK12/13 的高选择性,代表了进一步开发CDK12/13降解物作为TNBC患者的新靶向治疗的潜在先导分子。Octet® 验证7f来源化合物与CDK12 和 CDK13亲和力,Kd 值分别为 16.2 和 8.6 nM。Incucyte® 揭示化合物 7f可以显著抑制细胞增殖。本文用Incucyte® 和Octet® 分别在细胞水平和分子水平对小分子活性进行了验证。

降解剂 7f或DNA合成抑制剂(顺铂)均对MDA-MB-231细胞有中等程度的生长抑制作用。而7f与顺铂联合治疗可显著抑制细胞增殖。7f与PARP抑制剂奥拉帕尼(Olaparib)联合治疗也观察到类似的协同效应。


Octet® 检测7f来源化合物与CDK12和CDK13的亲和力,发现亲和力在nM。

TIL细胞杀伤:
Incucyte® 细胞杀伤实验好帮手

TIL(Tumor Infiltrating Lymphocytes,肿瘤浸润淋巴细胞)细胞疗法,主要依赖于肿瘤内部或附近的淋巴细胞已经具有识别和杀伤肿瘤细胞的能力,通过体外分离扩增并回输至病人体内,可以扩大免疫应答,加强肿瘤杀伤作用。但这种疗法的疗效可能会因 "容易耗尽 "而受到影响,体内持久性差而影响疗效。复旦大学附属妇产科医院发现抑制 PI3K-AKT 信号通路可以增强 TIL 的细胞毒性,可以改善当前基于 TIL 治疗实体瘤的前景的策略。Incucyte® 可以验证TIL杀伤能力。

2名宫颈癌患者的TIL细胞用 AKT和PIK3抑制剂处理,分别与Hele细胞和自体肿瘤细胞(ATC)共培养。与TIL对照组相比,具有AKT抑制作用的 TIL 对 HeLa 和 ATC 的细胞毒性显着增强。相比之下,PI3K抑制仅一位供体的TIL 对HeLa 细胞的细胞毒性有所改善,对 ATC 的杀伤没有显著影响。
