全基因敲除库（人）介绍

 

       全基因敲除库（人，gRNA pooled library in lentiCRISPRv2）购自Addgene（1000000048），是Human CRISPR Knockout Pooled Library (GeCKO v2)的一种，由张锋的团队构建（Improved vectors and genome-wide libraries for CRISPR screening. Nat Methods. 2014）。

       全基因敲除库（人）为慢病毒载体文库，载体骨架是lentiCRISPR v2（Addgene，52961）。lentiCRISPRv2能够表达sgRNA、Cas9和Puro抗性基因。慢病毒载体形式的文库能够提高感染与敲除效率，而且同时表达sgRNA和Cas9的单一载体能够应用于任何感兴趣的细胞类型，而不需要首先产生表达Cas9的细胞系。全基因敲除库（人）具有123411个sgRNA结构，靶向人类基因组中19050个编码基因及1864个miRNA。

 全基因敲除库（人）的使用方法

1、全基因敲除库（人）慢病毒包装。
2、慢病毒粒感染目标细胞，Puro筛选阳性细胞。
3、药物筛选或者辐射处理等，对照组不处理或者虚假处理。
4、提取基因组DNA，PCR扩增sgRNA的编码序列。
5、sgRNA的编码序列高通量测序。
6、比较对照组和实验组的高通量测序结果，筛选感兴趣的基因。
7、单独验证筛选出来的基因。
